Yes you can. You can use Houghi's website:

Map out your trip (start address, end address) using Google maps that is linked in Houghi's site. Make sure you zoom in to the start and stop locations and move them just a bit so Google maps will generate the lat/lon for these (you'll see the lat/lons added to the start and end address). When your trip is finished, click on 'link to this page'. Then copy that information (right-click copy) and paste it into the Houghi page and click 'make ITN'. If you've done everything right, you'll get an itinerary you can transfer to your TomTom.
Near the bottom of Houghi's page, there's an interesting video with some of the added features of Google maps... where you can drag your preferred route the way you want to go. But, you'll have to add some waypoints along that route to ensure that TomTom takes you that way.
There's also a program called TYRE (Trace Your Route Everywhere) that's free that also works pretty well.

This is awesome but since I live in Northern Virginia and Google maps refuses to take I66, this is pretty much useless to me. I need a way to do this with a map tool that will take I66.
Using Google maps, you can click on and drag the route to any road you want to travel. Did you view the video at the bottom of Houghi's page?
Not knowing where you live, I just created a route from Strasburg, VA to Shenandoah Farms, VA and verified this works. I was able to drag the route onto I-66 or onto any other road I wanted. Give it a try.

Yes, this is a well known problem and grief of those of us who live here. The problem is not all of 66 but only the east bound section inside the beltway. It's because of HOV restrictions and Google has made it impossible to drag a route onto it. For most cases where I am trying to force a route it is a situation where I want to take 66 toward DC. Most other cases I don't really need such a well defined itinerary. It is my understanding that there are other HOV roads that have the same problem.
